Node and leaf explants of Jatropha pandurifolia were cultured in Murashige and Skoog medium (MS) supplemented with plant growth regulators benzylaminopurine (BA), a-naphthaleneacetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D), alone or in combination. For the node explants, the treatments included MS + BA only at 2.5, 5.0, and 10.0 uM, MS + 10.0 uM BA + NAA or 2,4-D added singly at 1.25 uM, and MS + 10.0 uM BA + NAA and 2,4-D added in combination at 0.1 uM. In leaf explants, MS was supplemented only with NAA and 2,4-D, added alone or in combination at different concentrations. MS without plant growth regulator served as control. All node explants exposed to auxin and cytokinin growth regulators were observed to form calluses while all leaf explants except for the treatment with low concentration of NAA (2.5 uM) also formed calluses. Calluses were produced in leaf and node explants for 10 and 14 days inoculation in plant growth regulators, respectively. The most effective treatment for the production of callus in leaf and node explants were observed in the treatments of MS + 2.5 uM 2,4-D and MS + 10.0 uM BA + 1.25 uM NAA, respectively.
