Book Chapter Description

The in-situ nitrile hydratase/amidase (NHase/AMase) cascade enzymatic system is largely investigated in the literature for their biotechnological applications. This chapter compares the kinetic parameters of each catalyzed reaction (Km and activation energy) obtained in the presence of their respective substrate (acrylonitrile, propionitrile, benzonitrile, and 3-cyanopyridine for NHase and the corresponding amide for AMase). The enzyme stability is documented by long-term experiments performed in continuous stirred UF-membrane reactors. The inactivation constants and/or the activation energy of the inactivation process over a temperature range (5-60°C) are given. The effect of nitrile concentration on the activity and stability of the cascade enzymatic components is discussed.
