Book Chapter Description
This chapter discusses the isolation of sperm specific lactate dehydrogenase (LDH) from mouse, rabbit, human testis, and human spermatozoa. The isolation of sperm-specific antigens facilitates the analysis of immunologically induced infertility and of the physiological role of these unique gene products of the male gamete. One such protein is the lactate dehydrogenase C4 isozyme (LDH-C 4 ). This isozyme is distinct from the LDH isozymes found in other tissues. It is antigenic in both males and females, and the immune response to this isozyme can be manipulated to impair fertility. LDH-C 4 has been purified from testes of the rat by the conventional methods of salt fractionation, ion-exchange chromatography, and gel filtration. A generalized procedure for the isolation of LDH-C 4 can be based on an 8-(6-aminohexyl)-amino-5' AMP–Sepharose affinity gel and ion-exchange chromatography. The availability of this purified enzyme can facilitate studies of the role of the immune system in reproduction.
