This chapter illustrates C3a anaphylatoxin receptor, which is a G protein-coupled receptor of 482 amino acids characterized by the presence of an unusually large second extracellular domain. C3a receptor functions as the cell surface receptor for the anaphylatoxin C3a, the C-terminal 77 amino acid cleavage product of the α chain of C3, but not C3a-desArg. C3a and C4a are reported to act through the same receptor; however, this appears to be a unique property of the guineapig C3aR as C4a is not an agonist of the human or mouse C3aR. Transcript for the C3aR is widely distributed in peripheral tissues and the central nervous system. Using antibodies reactive with the second extracellular domain of the C3aR, expression is demonstrated on neutrophils, monocytes, eosinophils, astrocytes, neurons, and glial cells.
